target relevance
Protein names
Mitogen-activated protein kinase kinase kinase 14 (EC 2.7.11.25) (NF-kappa-beta-inducing kinase) (HsNIK) (Serine/threonine-protein kinase NIK)
Gene names
MAP3K14,MAP3K14 NIK
Protein family
Protein kinase superfamily, STE Ser/Thr protein kinase family, MAP kinase kinase ki
Mass

Function
FUNCTION: Lymphotoxin beta-activated kinase which seems to be exclusively involved in the activation of NF-kappa-B and its transcriptional activity. Promotes proteolytic processing of NFKB2/P100, which leads to activation of NF-kappa-B via the non-canonical pathway. Could act in a receptor-selective manner. {ECO:0000269|PubMed:15084608}.
Catalytic activity
CATALYTIC ACTIVITY: Reaction=ATP + L-seryl-[protein] = ADP + H(+) + O-phospho-L-seryl-[protein]; Xref=Rhea:RHEA:17989, Rhea:RHEA-COMP:9863, Rhea:RHEA-COMP:11604, ChEBI:CHEBI:15378, ChEBI:CHEBI:29999, ChEBI:CHEBI:30616, ChEBI:CHEBI:83421, ChEBI:CHEBI:456216; EC=2.7.11.25; CATALYTIC ACTIVITY: Reaction=ATP + L-threonyl-[protein] = ADP + H(+) + O-phospho-L-threonyl-[protein]; Xref=Rhea:RHEA:46608, Rhea:RHEA-COMP:11060, Rhea:RHEA-COMP:11605, ChEBI:CHEBI:15378, ChEBI:CHEBI:30013, ChEBI:CHEBI:30616, ChEBI:CHEBI:61977, ChEBI:CHEBI:456216; EC=2.7.11.25;
Subellular location
SUBCELLULAR LOCATION: Cytoplasm.
Tissues
TISSUE SPECIFICITY: Weakly expressed in testis, small intestine, spleen, thymus, peripheral blood leukocytes, prostate, ovary and colon. {ECO:0000269|PubMed:9020361}.
Structure
SUBUNIT: Interacts with TRAF2, TRAF5, TRAF6, IKKA and NFKB2/P100 (By similarity). Interacts with TRAF3 and PELI3. Interacts with NIBP; the interaction is direct. Interacts with ARRB1 and ARRB2. Interacts with GRB10. Interacts with ZFP91. Interacts with NLRP12; this interaction promotes proteasomal degradation of MAP3K14. Directly interacts with DDX3X (PubMed:30341167). Interacts (via C-terminus and kinase domain) with PPPC3A (via N-terminus) and PPP3CB (By similarity). {ECO:0000250, ECO:0000250|UniProtKB:Q9WUL6, ECO:0000269|PubMed:12853971, ECO:0000269|PubMed:12874243, ECO:0000269|PubMed:15084608, ECO:0000269|PubMed:15173580, ECO:0000269|PubMed:15951441, ECO:0000269|PubMed:17237370, ECO:0000269|PubMed:20682767, ECO:0000269|PubMed:30341167}.
Post-translational modification
PTM: Autophosphorylated. Phosphorylation at Thr-559 is required to activates its kinase activity and 'Lys-63'-linked polyubiquitination. Phosphorylated by CHUK/IKKA leading to MAP3K14 destabilization. {ECO:0000269|PubMed:20501937, ECO:0000269|PubMed:20682767}.; PTM: Ubiquitinated. Undergoes both 'Lys-48'- and 'Lys-63'-linked polyubiquitination. 'Lys-48'-linked polyubiquitination leads to its degradation by the proteasome, while 'Lys-63'-linked polyubiquitination stabilizes and activates it.
