FUNCTION: The exact function of MAP2 is unknown but MAPs may stabilize the microtubules against depolymerization. They also seem to have a stiffening effect on microtubules.

Structure
SUBUNIT: Interacts with KNDC1 (via KIND2); the interaction enhances MAP2 phosphorylation and localizes KNDC1 to dendrites. Interacts with DPYSL5 (PubMed:33894126). {ECO:0000250|UniProtKB:P20357, ECO:0000269|PubMed:33894126}.
Post-translational modification
PTM: Phosphorylated at serine residues in K-X-G-S motifs by MAP/microtubule affinity-regulating kinase (MARK1 or MARK2), causing detachment from microtubules, and their disassembly (By similarity). Isoform 2 is probably phosphorylated by PKA at Ser-323, Ser-354 and Ser-386 and by FYN at Tyr-67. Immunofluorescent analysis of adult rat hippocampus section stained with mouse mAb to MAP2A/B/C/D, 5291, dilution 1:5,000 in green, and chicken pAb to FOX2, dilution 1:2,000, in red. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45µM, and free-floating sections were stained with above antibodies. 5291 antibody labels all MAP2 protein isotypes expressed in neuronal perikarya and dendrites. The FOX2 antibody stains the nuclei of most neuronal cells.

Chromogenic immunostaining of a formalin fixed paraffin embedded human hippocampus section with mouse mAb to MAP2A/B/C/D, 5291, dilution 1:5,000, detected in DAB(brown) using the Vector Labs ImmPRESS method and reagents with citra buffer retrieval. Hematoxylin (blue) was used as the counterstain. In this image, the MAP2 antibody specifically labels neuronal cell bodies and projections. This antibody performs well in testing with both 4% PFA and standard NBF fixed tissues.

Western blot analysis of tissue and cell lysates using mouse mAb to MAP2C/D, 5291, dilution 1:5,000 in green, and chicken pAb to vimentin, 6886, dilution 1:5,000 in red. [1] protein standard (red), [2] rat whole brain lysate, [3] HeLa, [4] SH-SY5Y, [5] HEK293 and [6] NIH-3T3 cell lysates. A band at about 280kDa is full length intact MAP2A/2B isotypes while bands at about 70kDa represent MAP2C/D isotypes. Multiple bands in between are likely in vivo fragments of MAP2A/B. Only the SH-SY5Y cells, which have neuronal properties express MAP2 protein. The 6886 antibody binds to a single band at ~50kDa present in all preparations.
