NeuN, also known as Fox-3 or RBFOX3, is a well-established neuronal marker widely used in neuroscience research. It is a nuclear protein that is selectively expressed in post-mitotic neurons, making it an essential tool for identifying and characterizing neuronal populations in various tissues and experimental models. NeuN immunohistochemistry, facilitated by specific antibodies targeting NeuN, allows researchers to visualize and quantify neuronal cell bodies, providing valuable insights into neuronal distribution, density, and morphology. Immunofluorescent analysis of rat hippocampus section stained with chicken pAb to FOX3/NeuN, 1867, dilution 1:5,000 in red, and costained with rabbit pAb to IBA1 dilution 1:2,000, in green. Following transcardial perfusion with 4% paraformaldehyde, rat brain was post fixed for 24 hours, cut to 35µM, and free-floating sections were stained with the above antibodies. The FOX3/NeuN antibody stains the nuclei and distal perikarya of most neurons, while the IBA1 antibody specifically labels microglial cells.
