background | Interleukin-3 receptor subunit alpha, also known as IL-3 receptor subunit alpha, IL-3R-alpha, CD123, and IL3RA, is a single-pass type I membrane protein which belongs to the type I cytokine receptor family and Type 5 subfamily. The specific alpha subunit of the interleukin-3 receptor (IL-3Ralpha, CD123) is strongly expressed in various leukemic blasts and leukemic stem cells and seems to be an excellent target for the therapy of leukemias. |
molecular weight | The protein has a predicted MW of 36.12 kDa. Due to glycosylation, the protein migrates to 55-70 kDa based on Tris-Bis PAGE result. |
available size | 100 µg, 500 µg |
endotoxin | Less than 1EU per μg by the LAL method. |

Function Cell surface receptor for IL3 expressed on hematopoietic progenitor cells, monocytes and B-lymphocytes that controls the production and differentiation of hematopoietic progenitor cells into lineage-restricted cells (By similarity). Ligand stimulation rapidly induces hetrodimerization with IL3RB, phosphorylation and enzyme activity of effector proteins such as JAK2 and PI3K that play a role in signaling cell proliferation and differentiation (PubMed:10477686, PubMed:31990690). Activation of JAK2 leads to STAT5-mediated transcriptional program (PubMed:10376805). |
Subellular location [Isoform 1]: Cell membrane; Single-pass type I membrane protein. Note=Expressed on the cell surface.; [Isoform 2]: Endomembrane system; Single-pass type I membrane protein. Note=Mostly distributed inside the cells, except in the nuclei and is not transported to the cell surface. |
Structure Interacts with IL3. Heterodimer of an alpha and a beta subunit (PubMed:10477686). The beta subunit is common to the IL3, IL5 and GM-CSF receptors. |
Post-translational modification Ubiquitinated at Lys-357 by RNFT2 in response to IL3. Ubiquitination leads ligand-induced degradation by the proteasome. |
