background

Interleukin (IL)-12 and IL-23 belong to the IL-12 type family and are composite cytokines, consisting of the common β subunit p40 and the specific cytokine α subunit p35 and p19, respectively. IL-12 signals via the IL-12Rβ1·IL-12Rβ2 receptor complex, and IL-23 uses also IL-12Rβ1 but engages IL-23R as second receptor.

molecular weight

The protein has a predicted MW of 20.8 (Mouse IL-23 alpha)&35.8 (Mouse IL-12 beta) kDa. Due to glycosylation, the protein migrates to 22 (Mouse IL-23 alpha)&45-50 (Mouse IL-12 beta) kDa based on Tris-Bis PAGE result.

Function
Associates with IL12B to form the IL-23 interleukin, a heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T-cells and promotes production of pro-inflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis.; Associates with IL12B to form the pro-inflammatory cytokine IL-23 that plays different roles in innate and adaptive immunity (PubMed:11114383, PubMed:11390512, PubMed:16670770). Released by antigen-presenting cells such as dendritic cells or macrophages, binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R to activate JAK2 and TYK2 which then phosphorylate the receptor to form a docking site leading to the phosphorylation of STAT3 and STAT4 (By similarity). This process leads to activation of several pathways including p38 MAPK or NF-kappa-B and promotes the production of pro-inflammatory cytokines such as interleukin-17A/IL17A (PubMed:19900478). In turn, participates in the early and effective intracellular bacterial clearance (PubMed:16157683). Promotes the expansion and survival of T-helper 17 cells, a CD4-positive helper T-cell subset that produces IL-17, as well as other IL-17-producing cells (PubMed:16648837).
Subellular location
Secreted. Note=Secreted upon association with IL12B.
Tissues
Secreted by activated dendritic cells (at protein level). Detected in various tissues with higher expression in polarized Th1 cells and activated macrophages.
Structure
Heterodimer with IL12B; disulfide-linked (PubMed:11114383). The heterodimer is known as interleukin IL-23 (PubMed:11114383). Interacts with IL23R; this interaction enables recruitment of IL12RB1 (By similarity).
