background

CD74 (invariant MHC class II) regulates protein trafficking and is a receptor for macrophage migration inhibitory factor (MIF) and d-dopachrome tautomerase (d-DT/MIF-2). CD74 expression is increased in tubular cells and/or glomerular podocytes and parietal cells in human metabolic nephropathies, polycystic kidney disease, graft rejection and kidney cancer and in experimental diabetic nephropathy and glomerulonephritis.

molecular weight

The protein has a predicted MW of 26.5 kDa. Due to glycosylation, the protein migrates to 40-50 kDa based on Tris-Bis PAGE result.

available size

100 µg, 500 µg

endotoxin

Less than 1EU per μg by the LAL method.

Protein names
H-2 class II histocompatibility antigen gamma chain (Ia antigen-associated invariant chain) (Ii) (Ii chain) (MHC class II-associated invariant chain) (CD antigen CD74) [Cleaved into: Class-II-associated invariant chain peptide (CLIP)]
Gene names
Cd74,Cd74 Ii
Mass
10090Da
Function
Plays a critical role in MHC class II antigen processing by stabilizing peptide-free class II alpha/beta heterodimers in a complex soon after their synthesis and directing transport of the complex from the endoplasmic reticulum to compartments where peptide loading of class II takes place. Enhance also the stimulation of T-cell responses through interaction with CD44.; [Isoform Long]: Stabilizes the conformation of mature CTSL by binding to its active site and serving as a chaperone to help maintain a pool of mature enzyme in endocytic compartments and extracellular space of antigen-presenting cells (APCs).; [Class-II-associated invariant chain peptide]: Binds to the peptide-binding site of MHC class II alpha/beta heterodimers forming an alpha-beta-CLIP complex, thereby preventing the loading of antigenic peptides to the MHC class II complex until its release by HLA-DM in the endosome.
Catalytic activity
#N/A
Subellular location
[Isoform Long]: Late endosome. Lysosome .; Cell membrane ; Single-pass type II membrane protein. Endoplasmic reticulum membrane. Golgi apparatus, trans-Golgi network. Endosome. Lysosome. Secreted. Note=Transits through a number of intracellular compartments in the endocytic pathway. It can either undergo proteolysis or reach the cell membrane.
Tissues
[Isoform Long]: Expressed in thymus and lymph noodes (PubMed:8977190). Expressed by antigen-presenting cells (APCs) (PubMed:11483509, PubMed:12417635).; [Isoform Short]: Expressed in thymus and lymph noodes.
Structure
Nonamer composed of three alpha/beta/gamma heterotrimers. Interacts with CD44; this complex is essential for the MIF-induced signaling cascade that results in B cell survival.; [Isoform Long]: Interacts with the mature form of CTSL; the complex survive in neutral pH environment.
