background

Hepatocyte growth factor (HGF) is an important component of the pathophysiology of IR, with increased levels in most common IR conditions, including obesity. HGF has a role in the metabolic flux of glucose in different insulin sensitive cell types.

molecular weight

The HGF single chain can be processed into the active form of disulfide-linked heterodimer of α and β chain. The protein has a predicted MW of 79.6 kDa (after cleavage of the signal peptide). Due to glycosylation, the protein migrates to 60-65 kDa and 30-40 kDa corresponding to α chain and β chain based on Bis-Tris PAGE result.

target relevance
Hepatocyte growth factor (HGF) is an important component of the pathophysiology of IR, with increased levels in most common IR conditions, including obesity. HGF has a role in the metabolic flux of glucose in different insulin sensitive cell types.
Protein names
Hepatocyte growth factor (Hepatopoietin-A) (Scatter factor) (SF) [Cleaved into: Hepatocyte growth factor alpha chain; Hepatocyte growth factor beta chain]
Gene names
HGF,HGF HPTA
Protein family
Peptidase S1 family, Plasminogen subfamily
Mass
9606Da
Function
Potent mitogen for mature parenchymal hepatocyte cells, seems to be a hepatotrophic factor, and acts as a growth factor for a broad spectrum of tissues and cell types (PubMed:20624990). Activating ligand for the receptor tyrosine kinase MET by binding to it and promoting its dimerization (PubMed:15167892, PubMed:20977675). Activates MAPK signaling following TMPRSS13 cleavage and activation (PubMed:20977675).
Catalytic activity
#N/A
Structure
Dimer of an alpha chain and a beta chain linked by a disulfide bond. Interacts with SRPX2; the interaction increases HGF mitogenic activity.
Post-translational modification
The single-chain precursor undergoes proteolytic processing by TMPRSS13 resulting in an active two-chain form.
