background

B7-1 and B7-2 are homologous costimulatory ligands expressed on the surface of antigen presenting cells (APCs). Binding of these molecules to the T cell costimulatory receptors, CD28 and CTLA-4, is essential for the activation and regulation of T cell immunity. B7-1 and B7-2 do not form hetero-oligomers, underscoring the biological relevance of dimeric and monomeric state of B7-1 and B7-2, respectively.

molecular weight

The protein has a predicted MW of 28.2 kDa. Due to glycosylation, the protein migrates to 55-70 kDa based on Tris-Bis PAGE result.

Function
FUNCTION: Receptor involved in the costimulatory signal essential for T-lymphocyte proliferation and interleukin-2 production, by binding CD28 or CTLA-4. May play a critical role in the early events of T-cell activation and costimulation of naive T-cells, such as deciding between immunity and anergy that is made by T-cells within 24 hours after activation (PubMed:7527824). Also involved in the regulation of B cells function, plays a role in regulating the level of IgG(1) produced. Upon CD40 engagement, activates NF-kappa-B signaling pathway via phospholipase C and protein kinase C activation (By similarity). {ECO:0000250|UniProtKB:P42082, ECO:0000269|PubMed:7527824}.; FUNCTION: [Isoform 2]: Interferes with the formation of CD86 clusters, and thus acts as a negative regulator of T-cell activation. {ECO:0000269|PubMed:7527824}.; FUNCTION: (Microbial infection) Acts as a receptor for adenovirus subgroup B. {ECO:0000269|PubMed:16920215}.
Subellular location
SUBCELLULAR LOCATION: Cell membrane; Single-pass type I membrane protein.
Tissues
TISSUE SPECIFICITY: Expressed by activated B-lymphocytes and monocytes.
Structure
SUBUNIT: Homodimer. Interacts with MARCH8. Interacts (via cytoplasmic domain) with PHB1 and PHB2; the interactions increases after priming with CD40 (By similarity). {ECO:0000250|UniProtKB:P42082, ECO:0000269|PubMed:11279501, ECO:0000269|PubMed:12582153, ECO:0000269|PubMed:12606712, ECO:0000305}.; SUBUNIT: (Microbial infection) Interacts with adenovirus subgroup b fiber protein. {ECO:0000269|PubMed:16920215}.; SUBUNIT: (Microbial infection) Interacts with Orthopoxvirus OPG038/M2 protein, inhibiting the interaction with CTLA4 and CD28. {ECO:0000269|PubMed:30918073}.
Post-translational modification
PTM: Polyubiquitinated; which is promoted by MARCH8 and results in endocytosis and lysosomal degradation. {ECO:0000269|PubMed:12582153}.
