background

B7-H5, also known as VISTA, B7H5, Dies1, SISP1 and C10orf54, is a 55-65 kDa member of the Ig superfamily. It is a transmembrane molecule expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces.

molecular weight

The protein has a predicted MW of 21.04 kDa. Due to glycosylation, the protein migrates to 40-60 kDa based on Tris-Bis PAGE result.

target relevance
B7-H5, also known as VISTA , B7H5, Dies1, SISP1 and C10orf54, is a 55-65 kDa member of the Ig superfamily. It is a transmembrane molecule expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces.
Protein names
V-type immunoglobulin domain-containing suppressor of T-cell activation (Platelet receptor Gi24) (Stress-induced secreted protein-1) (Sisp-1) (V-set domain-containing immunoregulatory receptor) (V-set immunoregulatory receptor)
Mass
33908Da
Function
Immunoregulatory receptor which inhibits the T-cell response (PubMed:24691993). May promote differentiation of embryonic stem cells, by inhibiting BMP4 signaling (By similarity). May stimulate MMP14-mediated MMP2 activation (PubMed:20666777). {ECO:0000250|UniProtKB:Q9D659, ECO:0000269|PubMed:20666777, ECO:0000269|PubMed:24691993}.
Subellular location
Cell membrane {ECO:0000269|PubMed:20666777, ECO:0000269|PubMed:24691993}; Single-pass type I membrane protein {ECO:0000255}.
Tissues
Expressed in spleen. Detected on a number of myeloid cells including CD11b monocytes, CD66b+ neutrophils, at low levels on CD4+ and CD8+ T-cells, and in a subset of NK cells. Not detected on B cells (at protein level). Expressed at high levels in placenta, spleen, plasma blood leukocytes, and lung. Expressed at moderate levels in lymph node, bone marrow, fat, uterus, and trachea. Has low expression levels in other tissues. {ECO:0000269|PubMed:24691993}.
Post-translational modification
At the cell surface, may be cleaved by MMP14. {ECO:0000269|PubMed:20666777}.; N-glycosylated. {ECO:0000250|UniProtKB:Q9D659}.
