Vantictumab Vantictumab biologic drug binds Homo sapiens FZD5,Homo sapiens FZD10
Homo sapiens FZD5 Frizzled-5
Protein names Frizzled-5
Alternative names FzE5
Gene names FZD5
Protein family Belongs to the G-protein coupled receptor Fz/Smo family
Function Receptor for Wnt proteins (PubMed:10097073, PubMed:20530549, PubMed:26908622, PubMed:9054360). Functions in the canonical Wnt/beta-catenin signaling pathway. In vitro activates WNT2, WNT10B, WNT5A, but not WNT2B or WNT4 signaling (By similarity). In neurons, activation by WNT7A promotes formation of synapses (PubMed:20530549). May be involved in transduction and intercellular transmission of polarity information during tissue morphogenesis and/or in differentiated tissues (Probable). Plays a role in yolk sac angiogenesis and in placental vascularization (By similarity). Plays a role in ocular development (PubMed:26908622)
Structure Binding of unsaturated fatty acid molecules (via FZ domain) promotes homodimerization (PubMed:28377511). Interacts with WNT2B (PubMed:12490564). Interacts with WNT3A (PubMed:26908622). Interacts with WNT7A (PubMed:18230341, PubMed:20530549, PubMed:26908622). Interacts with GOPC (By similarity)
Post-translational modification Ubiquitinated by RNF43 and ZNRF3, leading to its degradation by the proteasome
Involvement in disease Microphthalmia/Coloboma 11 A form of colobomatous microphthalmia, a disorder of eye formation, ranging from small size of a single eye to complete bilateral absence of ocular tissues. Ocular abnormalities like coloboma, opacities of the cornea and lens, scaring of the retina and choroid, and other abnormalities may also be present. Ocular colobomas are a set of malformations resulting from abnormal morphogenesis of the optic cup and stalk, and the fusion of the fetal fissure (optic fissure). MCOPCB11 is an autosomal dominant form with incomplete penetrance.
Protein family Belongs to the G-protein coupled receptor Fz/Smo family
Function Receptor for Wnt proteins. Functions in the canonical Wnt/beta-catenin signaling pathway (By similarity). The canonical Wnt/beta-catenin signaling pathway leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes. A second signaling pathway involving PKC and calcium fluxes has been seen for some family members, but it is not yet clear if it represents a distinct pathway or if it can be integrated in the canonical pathway, as PKC seems to be required for Wnt-mediated inactivation of GSK-3 kinase. Both pathways seem to involve interactions with G-proteins. May be involved in transduction and intercellular transmission of polarity information during tissue morphogenesis and/or in differentiated tissues (Probable)
Subcellular location Cell membrane
Structure Interacts with WNT7B (By similarity). Interacts with MYOC (PubMed:19188438)
Post-translational modification Ubiquitinated by ZNRF3, leading to its degradation by the proteasome
Protein family Belongs to the G-protein coupled receptor Fz/Smo family
Function Receptor for Wnt proteins. Most of frizzled receptors are coupled to the beta-catenin canonical signaling pathway, which leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes. May be involved in transduction and intercellular transmission of polarity information during tissue morphogenesis and/or in differentiated tissues. Together with FZD3, is involved in the neural tube closure and plays a role in the regulation of the establishment of planar cell polarity (PCP), particularly in the orientation of asymmetric bundles of stereocilia on the apical faces of a subset of auditory and vestibular sensory cells located in the inner ear (By similarity)
Post-translational modification Ubiquitinated by ZNRF3, leading to its degradation by the proteasome
Involvement in disease Nail disorder, non-syndromic congenital, 1 An autosomal recessive nail disorder characterized by a variable degree of onychauxis (thick nails), hyponychia, and onycholysis of all nails, with claw-shaped fingernails in some individuals. No other anomalies of ectodermal tissues, including hair, teeth, sweat glands, or skin, are noted, and individuals with dysplastic nails have normal hearing and normal psychomotor development.
