background | Latent TGF beta 1 cDNA encodes a 390 amino acid precursor that contains a 29 aa signal peptide and a 361 aa proprotein. A furin‑like convertase processes the proprotein to generate an N‑terminal 249 aa latency‑associated peptide (LAP) and a C‑terminal 112 aa mature TGF‑ beta 1. Disulfide‑linked homodimers of LAP and TGF‑ beta 1 remain non‑covalently associated after secretion, forming the small latent TGF‑ beta 1 complex. |
molecular weight | The protein has a predicted MW of 31.4/12.8 kDa. Due to glycosylation, the protein migrates to 40-50/14-15 kDa based on Tris-Bis PAGE result. |
available size | 100 µg, 500 µg |

Function FUNCTION: Required to maintain the Transforming growth factor beta-1 (TGF-beta-1) chain in a latent state during storage in extracellular matrix. Associates non-covalently with TGF-beta-1 and regulates its activation via interaction with 'milieu molecules', such as LTBP1, LRRC32/GARP and LRRC33/NRROS, that control activation of TGF-beta-1. Interaction with LRRC33/NRROS regulates activation of TGF-beta-1 in macrophages and microglia. Interaction with LRRC32/GARP controls activation of TGF-beta-1 on the surface of activated regulatory T-cells (Tregs). Interaction with integrins (ITGAV:ITGB6 or ITGAV:ITGB8) results in distortion of the Latency-associated peptide chain and subsequent release of the active TGF-beta-1. {ECO:0000256|ARBA:ARBA00003201}.; FUNCTION: Transforming growth factor beta-1 proprotein: Precursor of the Latency-associated peptide (LAP) and Transforming growth factor beta-1 (TGF-beta-1) chains, which constitute the regulatory and active subunit of TGF-beta-1, respectively. {ECO:0000256|ARBA:ARBA00002007}. |
Subellular location SUBCELLULAR LOCATION: Secreted, extracellular space, extracellular matrix {ECO:0000256|ARBA:ARBA00004498}. |
Structure SUBUNIT: Homodimer; disulfide-linked. Interacts with TGF-beta receptors (TGFBR1 and TGFBR2), leading to signal transduction. {ECO:0000256|ARBA:ARBA00046998}. |
