background

CSF-1, also known as M-CSF, is a four alpha -helical-bundle cytokine that is the primary regulator of macrophage survival, proliferation and differentiation. CSF-1 is also essential for the survival and proliferation of osteoclast progenitors.CSF-1 is cytokine that plays an essential role in the regulation of survival, proliferation and differentiation of hematopoietic precursor cells, especially mononuclear phagocytes, such as macrophages and monocytes. Promotes the release of proinflammatory chemokines, and thereby plays an important role in innate immunity and in inflammatory processes. This mutation leads to a deletion of exons 2-7 of the EGFR gene and renders the mutant receptor incapable of binding any known ligand.

molecular weight

The protein has a predicted MW of 26.26 kDa. Due to glycosylation, the protein migrates to 45-55 kDa based on Tris-Bis PAGE result.

Function
Cytokine that plays an essential role in the regulation of survival, proliferation and differentiation of hematopoietic precursor cells, especially mononuclear phagocytes, such as macrophages and monocytes. Promotes the release of pro-inflammatory chemokines, and thereby plays an important role in innate immunity and in inflammatory processes. Plays an important role in the regulation of osteoclast proliferation and differentiation, the regulation of bone resorption, and is required for normal bone development. Required for normal male and female fertility. Promotes reorganization of the actin cytoskeleton, regulates formation of membrane ruffles, cell adhesion and cell migration. Plays a role in lipoprotein clearance.
Subellular location
Cell membrane ; Single-pass type I membrane protein .; [Processed macrophage colony-stimulating factor 1]: Secreted, extracellular space.
Structure
Homodimer or heterodimer; disulfide-linked (PubMed:1531650, PubMed:3264877, PubMed:8422357). Likely to exist in multiple forms: homodimer consisting of 2 identical 150-200 kDa proteoglycan subunits, heterodimer consisting of a 150-200 kDa proteoglycan subunit and a truncated 43 kDa subunit, and homodimer consisting of 2 identical 43 kDa subunits (PubMed:8051056). Interacts with CSF1R (PubMed:20504948).
Post-translational modification
N-glycosylated.; [Isoform 1]: N-glycosylated.; [Isoform 3]: N-glycosylated.; O-glycosylated; contains chondroitin sulfate (PubMed:1531650, PubMed:8051056). O-glycosylated with core 1 or possibly core 8 glycans (PubMed:22171320, PubMed:23234360, PubMed:3264877).; [Isoform 1]: O-glycosylated.
