target relevance
ULBP2 expression was however linked to expression of mature CD57() NK cells. In particular, expression of ULBP2 was strongest on those NK cells that had evidence of recent activation and proliferation. We suggest that ULBP2 could be used to identify recently activated "mature" NK cells.
Protein names
UL16-binding protein 2 (ALCAN-alpha) (NKG2D ligand 2) (N2DL-2) (NKG2DL2) (Retinoic acid early transcript 1H)
Gene names
ULBP2,ULBP2 N2DL2 RAET1H UNQ463/PRO791
Protein family
MHC class I family

9606Da
Function
FUNCTION: Binds and activates the KLRK1/NKG2D receptor, mediating natural killer cell cytotoxicity. {ECO:0000269|PubMed:11777960}.
Subellular location
SUBCELLULAR LOCATION: Cell membrane {ECO:0000269|PubMed:12782710}; Lipid-anchor, GPI-anchor {ECO:0000269|PubMed:12782710}. Endoplasmic reticulum {ECO:0000269|PubMed:12782710}. Secreted {ECO:0000269|PubMed:11444831}. Note=In CMV-infected fibroblasts, detected in the endoplasmic reticulum/cis-Golgi. {ECO:0000269|PubMed:12782710}.
Tissues
TISSUE SPECIFICITY: Expressed in various types of cancer cell lines and in the fetus, but not in normal tissues. {ECO:0000269|PubMed:11444831}.
Structure
SUBUNIT: Interacts with KLRK1/NKG2D (PubMed:11777960). Does not bind to beta2-microglobulin (PubMed:12782710). {ECO:0000269|PubMed:11777960, ECO:0000269|PubMed:12782710}.; SUBUNIT: (Microbial infection) In CMV-infected cells, interacts with the viral glycoprotein UL16; this interaction causes ULBP2 retention in the endoplasmic reticulum and cis-Golgi and prevents binding to and activation of KLRK1/NKG2D, providing CMV with an immune evasion mechanism. {ECO:0000269|PubMed:12782710}.

Data

ELISA with Biotinylated Human ULBP-2 Protein
Immobilized Biotinylated Human ULBP-2, His Tag at 1µg/ml (100µl/Well) on the streptavidin precoated plate (5µg/ml). Dose response curve for Human NKG2D, hFc Tag with the EC50 of 0.25µg/ml determined by ELISA.
Binding assay with Biotinylated Human ULBP-2 Protein
Serial dilutions of Anti-NKG2D Antibody, hFc Tag were added into Biotinylated Human ULBP-2, His Tag : Human NKG2D, hFc Tag binding reactioins. The half maximal inhibitiory concentration (IC50) is 45.2ng/ml.
HPLC of Biotinylated Human ULBP-2 Protein
The purity of Biotinylated Human ULBP-2 is greater than 95% as determined by SEC-HPLC.
SDS-PAGE gel of Biotinylated Human ULBP-2 Protein
